The Effect of Polyamine upon Sampenous Nerve Regeneration in the Rat


Abstract
Polyamine has been known to devote to cellular proliferation and growth in peripheral nerve regeneration and this study has been done to investigate the effect of polyamine and the interaction of nerve growth factor which is known as nerve cell
growith
promoting substance. Saphenous nerve of the rat has been injured with hemostat and polyamine, anti-nerve growth factor and DFMO have been injected. After certain period, axon diameter and myelin thickness have been measured in proximal and distl
portion
of the injury site of saphenous nerve. Followings are the results.
1) Axon diameter and myelin thickness decreased until 2 months and increased after 3 months after injury, however, they did not recovered to normal size even after 7 months after injury.
2) In 1 month after polymine treatment, proximal axon diameter increased and they were larger than normal group, however, 7 months group didn't show the increment. Myelin thickness of polyamine group showed no difference with normal control
group,
while they were increased compared to the experimental control group, In distal portion, axon diameter and myelin thickness were smaller than the normal control, while they were larger than experimental control group.
3) In polyamine and anti-NGF group, axon diameter of the proximal portion were increased a little bit in the early period and later no changes has been shown. However, myelin thickness were decreased in a large amount and they were similar to
experimental control group. In distal portion, axon diameter and myelin thickness were smaller than the normal control, while they were larger than experimental control group.
4) In DFMO group, axon diameter and myelin thickness of the proximal portion decreased in the early period and they were recovered after that. In 7 months group, they were almost same as normal control group. In the distal portion, axon diameter
an
myelin thickness were deceased and they were never recovered nerve, the shape of the nerve axon was after 7 months.
5) In unmyelinated nerve, the shape of the nerve axon was very irregular in experimental control group. Also, Schwann cell has the morphology of macrophages. In polyamine group, the diameters of nerve axon were increased through all period. In
polyamine-anti-NGF group, axons with various sizes were appeared and especially, small axons were obaerved in a large amount.
Axon diameter were increased in polyamine group and polyamine-anti-NGF group both in proximal and distal group. It could be assumed that plyamine has the effect on the regeneration of both proximal and distal protion of the nerve injury. Myelin
thickness was increased in polyamine group, however, they were not changed polyamine-anti-NGF group. With these results, it could be assumed that NGF may have more effect in myelin thickness in regeneration. With the above results, it could be
concluded
that polyamine has the effects in the early period of nerve regeneration.
